So how do you differentiate a Pathan then - only those who can speak Pushto

Yes , a pashtun is one whose mother tongue is pashto. Those who speak pashto but do not follow pashtunwali are considered "kamaya" or "kamasal".

Lolz. Many Punjabs live in Peshawar and can speak pashto.

Would that then make them Pathans? :rolleyes:

If they are identifying themeselves as punjabis and their mother tongue is still punjabi then they are ofcourse punjabis.
